Yes, dear Avi, I always use manual focus; autofocus is useless when I'm dealing with highly magnified macros and extreme shallow DoF.

Francisco, this is quite amazing! Well done composition! You appear to have two subjects, the blue flower by virtue of position and the foreground bud by virtue of point of focus. It is all in balance and seems to work very well!
